Immune-related Gastrointestinal Toxicities
Overview
As an independent investigator, Dr. Dougan has focused his efforts on understanding the etiology and immune mechanisms underlying the immune-related adverse events (irAEs) resulting from cancer immunotherapy, a window into the endogenous function of the immune receptors targeted by these therapies. His research dovetails with his clinical work, where he specializes in treating gastrointestinal (GI) irAEs resulting from “checkpoint” receptor blockade, which are often treatment limiting. Management relies on systemic corticosteroids, which are likely to have deleterious effects on antitumor responses. His goal is to use a detailed mechanistic understanding of gastrointestinal irAEs to develop novel treatment strategies that do not interfere with antitumor immunity, as well as to understand on a more fundamental level, how individual immune receptors regulate immune homeostasis in the GI tract.

Infliximab for the Treatment of Immune Checkpoint Inhibitor (ICI) Colitis
A randomized, open-label clinical trial to evaluate the efficacy and safety of the medication infliximab (Remicade, Inflectra) compared to steroids for the treatment of ICI colitis in patients with stage III/IV skin cancer.
PD-1 Antibody Therapy + Infliximab for Metastatic Melanoma
A randomized, double-blind clinical trial testing the safety and effectiveness of the investigational combination of anti-Programmed Death (PD)-1 antibody therapy with or without LAG-3 inhibition (pembrolizumab or nivolumab+relatlimab) and infliximab in treating metastatic melanoma.
Gastrointestinal Complications in Cancer Immunotherapy Patients
A biorepository study that uses biological samples (blood, stool, and intestinal biopsies) and related health information collected from different groups of patients to increase understanding about the causes of gastrointestinal complications in patients receiving immunotherapy, as well as the factors that may influence the course of these complications over time.
ICI Colitis Data Repository
A data repository focused on creating a central location for all data surrounding patients who experience GI toxicities due to ICIs.
